Abstract

Surunkali virusli gepatit B (HBV) O‘zbekiston o‘rtacha–yuqori endemiklik darajasiga ega hudud bo‘lib, mahalliy populyatsiyada D genotipi ustunlik qiladi. Virusning yuqori mutatsiya tezligi molekulyar-genetik diagnostikaning sezuvchanligiga ta’sir ko‘rsatadi va soxta-manfiy natijalar xavfini oshiradi. HBV genomining real vaqt PZR (qPCR) diagnostikasi uchun eng barqaror nukleotid hududlarini aniqlash va ushbu hududlarga yo‘naltirilgan praymer/zond to‘plamlarini in silico baholash. 170 ta genom ketma-ketligi (GenBank bazasidan 160 ta va O‘zbekiston hududiga mansub 10 ta izolyat) UGENE (v.49.0) muhitida MAFFT algoritmi yordamida ko‘p tomonlama tekislandi. Har bir pozitsiyaning barqarorligi Shennon entropiyasi (H) va polimorfizm chastotasi orqali baholandi (konservativlik mezoni: H ≤ 0.02 va variabellik ≤ 2%). Praymer va zondlarning birikish koordinatalari referens genom (GenBank NC_003977.2) bo‘yicha aniqlandi. Eng yuqori barqarorlikka ega hududlar Core (C) geni (1901–2452 nt, konservativlik 99.8–100%) va S genining N-terminal sohasi (155–369 nt, 98.1%) ekanligi aniqlandi. Core genining 3′- sohasiga (2338–2448 nt) yo‘naltirilgan GenLab HBC03 praymer/zond to‘plami o‘rganilgan D genotipi subgenotiplari (D1, D2, D3) bo‘yicha to‘liq gomologiyani namoyon etdi va birlamchi diagnostik nishon sifatida tavsiya etiladi. S genining N-terminal sohasiga (248–344 nt) loyihalangan GenLab HBS01 to‘plamining zondi A, B, C, D va E genotiplari bo‘yicha 100% konservativlikni ko‘rsatdi. Aksincha, S genining «a»-determinanta sohasi (524–595 nt) hamda bazal kor-promotor va pre-kor sohalari mutatsiyalarga moyilligi sababli birlamchi nishon sifatida tavsiya etilmaydi. Olingan natijalar mahalliy genetik landshaftga moslashtirilgan «in-house» qPCR test-tizimlarini yaratish uchun ilmiy asos bo‘lib xizmat qiladi.
