Abstract
Ushbu innovatsion metabolik muhandisligi tadqiqotida ilk bor Artemisia annua L. o‘simligining cADS geni yordamida g‘o‘za (G. hirsutum) o‘simligida artemizinin va arteannuin B heterologik biosintezini yo‘lga qo‘yishga qaratilgan tahlil natijalari keltirilgan. In silico 3D-strukturaviy tahlil AaADS va endogen GhCAD oqsillari o‘rtasida yuqori fazoviy gomologiyani (RMSD = 0,941 Å) ko‘rsatib, ekzogen fermentning funksional faolligini asosladi. Overekspression pBINPLUS:Aa-cADS vektori A. tumefaciens vositasida transformatsiya qilinib, somatik embriogenez orqali biotexnologik g‘o‘za liniyalari olindi. Molekulyar tahlillarda cADS genining g‘o‘za genomiga integratsiyasi va ekspressiyasi tasdiqlandi. UHPLC-QTOF-MS usulida o‘tkazilgan metabolomik tahlillar orqali transformant liniyalarda maqsadli metabolitlar (artemizinin va arteannuin B) sintezi aniqlandi.References
